New hope for tumor immunotherapy: the macrophage-related “do not eat me” signaling pathway

چکیده
The “do not eat me” signaling pathway is extremely active in tumor cells, providing a means for these cells to elude macrophage phagocytosis and escape immune surveillance. Representative markers of this pathway, such as CD47 CD24, are highly expressed numerous tumors. interaction SIRPα with reduces the accumulation non-myosin ⅡA on cell membrane. combination CD24 Siglec10 ultimately leads recruitment SHP-1 or SHP-2 reduce signal transduction. Both them weaken ability macrophages engulf cells. Blocking mutual recognition between CD47-SIRPα CD24-Siglec10 using large molecular proteins small drugs represents promising avenue immunotherapy. Doing so can inhibit transduction enhance clearance rates cancer In paper, we summarize characteristics that affect via classical molecule drugs, which target pathways, pathways. We expect it will offer insight into development new centered blocking pathway.

In 1890, Coley observed that cancer patients who developed infections had smaller tumors. From this, he developed Coley’s toxin and treated tumors with injections of infectious materials. In 1960s, Mahaley used monoclonal antibodies to treat central nervous system(CNS) tumors, that research and clinical investigations in brain tumor immunotherapy became a serious undertaking. ...
متن کاملBeginnings of a good apoptotic meal: the find-me and eat-me signaling pathways.
Prompt and efficient clearance of apoptotic cells is necessary to prevent secondary necrosis of dying cells and to avoid immune responses to autoantigens. Recent studies have shed light on how apoptotic cells through soluble "find-me" signals advertise their presence to phagocytes at the earliest stages of cell death. Phagocytes sense the find-me signal gradient, and in turn the presence of dyi...

Background: Bacterial DNA has immunostimulatory effects on different types of immune cells such as dendritic cells (DCs). Application of DCs as a cellular adjuvant represents a promising approach in the immunotherapy of infectious disease and cancers. Objectives: To investigate the effect of tumor antigen pulsed DCs in the presence of CpG-1826 in treatment of a murine model of cancer. Methods: ...
متن کاملEstimation of the Domain of Attraction of Free Tumor Equilibrium Point for Perturbed Tumor Immunotherapy Model
In this paper, we are going to estimate the domain of attraction of tumor-free equilibrium points in a perturbed cancer tumor model describing the tumor-immune system competition dynamics. The proposed method is based on an optimization problem solution for a chosen Lyapunov function that can be casted in terms of Linear Matrix Inequalities constraint and Taylor expansion of nonlinear terms. We...
متن کاملI-43: Expression Profile of Macrophage Migration Inhibitory Factor (MIF) Signaling Pathway as A Potentional Biomarker in Pathophysiology of Endometriosis
Background MIF via its receptor, CD74, initiates a signaling cascade that leads to proliferation and survival of cells. Also, MIF binding to CD74 activates p38 signaling pathways that lead to positive effect on the expression of COX-2. The aim of this study was to evaluate the gene expression profile of MIF, CD74 and COX-2 in normal, ectopic and eutopic endometrium during menstrual cycle. The e...
